I wrote this poem in December 2013 in the morning after sitting on the veranda of our home in the Blue Mountains in New South Wales Australia. The morning had been cold and raining, but that does not put a damper on my inspiration. The music in my head had been banging non-stop ever since I had arrived home from a trip to the spiritual shop with my treasures from The Timeshare. Intracellular pH (pHi) was measured in single, isolated, glucose-stimulated brown adipocytes by means of the weakly fluorescent 2,7-bis(carboxyethyl)-5,6-carboxyfluorescein (BCECF). The pHi of unstimulated brown adipocytes (pHi = 7.29 +/- 0.01, n = 24) was less negative than that of intact perfused tissues (pHi = 7.42 +/- 0.01, n = 9) and contained an alkaline-resistant component (pHi = 6.93 +/- 0.01, n = 6) that was rapidly dissipated upon cell stimulation. External alkalization of the cell interior caused a marked, transient acidification of pHi with a time course similar to that of the rise in membrane potential (Vm) and similar to that of pHi recovery from intracellular acidification. It was concluded that the high electrical negativity of the brown adipocyte interior, as compared to the extracellular space, is related to the acidification of its intracellular space upon stimulation. The kinetics of recovery from pHi acidification were similar to those of Vm, suggesting that an intrinsic electrical mechanism is involved. External acidification had only minor effects on the rate of pHi recovery. Whereas the intracellular acidification was sensitive to extracellular Na+ activity and was reduced in Cl- -free media, pHi recovery was insensitive to alterations in Na+
